This condition leads to progressive photoreceptor degeneration, causing symptoms such as difficulty seeing in low light, poor adaptation to changes in lighting, light sensitivity, and progressive loss of peripheral visual field. Although these mutations are rare and the disease affects approximately one in 40,000 young males, XLRP currently impacts more than <strong data-start=\"1063\" data-end=\"1082\">17,000 patients<\/strong> in the US and the EU5 countries (Italy, France, Germany, Spain, and the United Kingdom). Unfortunately, <strong data-start=\"1187\" data-end=\"1237\">no therapeutic options are currently available<\/strong>, and both patients and specialists have long awaited a new opportunity for treatment through <strong data-start=\"1331\" data-end=\"1347\">gene therapy<\/strong>.<\/p>\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-2f4b65c elementor-widget elementor-widget-text-editor\" data-id=\"2f4b65c\" data-element_type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t<p data-start=\"1350\" data-end=\"1892\">Gene therapy aims to restore lost cellular function caused by a genetic defect by delivering a \u201chealthy\u201d copy of the mutated gene. Currently, the <strong data-start=\"1496\" data-end=\"1551\">only approved gene therapy for retinitis pigmentosa<\/strong> is Spark Therapeutics\u2019 <strong data-start=\"1575\" data-end=\"1611\">Luxturna (voretigene neparvovec)<\/strong>, which is indicated only for RP caused by <strong data-start=\"1654\" data-end=\"1695\">biallelic mutations in the RPE65 gene<\/strong>. A <strong data-start=\"1699\" data-end=\"1758\">specific gene therapy for XLRP caused by RPGR mutations<\/strong> has been developed by <strong data-start=\"1781\" data-end=\"1804\">Beacon Therapeutics<\/strong>, which recently announced the launch of a <strong data-start=\"1847\" data-end=\"1875\">Phase III clinical trial<\/strong> named <strong data-start=\"1882\" data-end=\"1891\">VISTA<\/strong>.<\/p>\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-2d93750 elementor-widget elementor-widget-text-editor\" data-id=\"2d93750\" data-element_type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t<p data-start=\"1894\" data-end=\"2375\">The new gene therapy for X-linked retinitis pigmentosa uses <strong data-start=\"1954\" data-end=\"1996\">AGTC-501 (laruparetigene zavoparvovec)<\/strong>, a recombinant adeno-associated viral vector delivered through a <strong data-start=\"2062\" data-end=\"2095\">single intravitreal injection<\/strong>. This therapy enables retinal cells to resume production of the full RPGR protein. Due to these characteristics, researchers believe AGTC-501 can <strong data-start=\"2242\" data-end=\"2303\">preserve the function of both rod and cone photoreceptors<\/strong>, significantly improving quality of life for patients affected by XLRP.<\/p>\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-e1f7341 elementor-widget elementor-widget-text-editor\" data-id=\"e1f7341\" data-element_type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t<p data-start=\"2377\" data-end=\"3156\">Previous clinical trials of AGTC-501 (<strong data-start=\"2415\" data-end=\"2426\">HORIZON<\/strong> and <strong data-start=\"2431\" data-end=\"2442\">SKYLINE<\/strong>) have shown <strong data-start=\"2455\" data-end=\"2476\">excellent results<\/strong> in terms of <strong data-start=\"2489\" data-end=\"2512\">safety and efficacy<\/strong>, demonstrating good tolerability, no clinically significant adverse events, and improvements in retinal sensitivity and visual function. <strong data-start=\"2650\" data-end=\"2659\">VISTA<\/strong>, a randomized, double-blind, multicenter <strong data-start=\"2701\" data-end=\"2720\">Phase III trial<\/strong>, is designed to evaluate the <strong data-start=\"2750\" data-end=\"2788\">safety, efficacy, and tolerability<\/strong> of a <strong data-start=\"2794\" data-end=\"2825\">single subretinal injection<\/strong> of AGTC-501 at two dosage levels compared to an untreated control group. The study will enroll approximately <strong data-start=\"2935\" data-end=\"2955\">75 male patients<\/strong> between the ages of 12 and 50, all affected by X-linked retinitis pigmentosa caused by <strong data-start=\"3043\" data-end=\"3073\">mutations in the RPGR gene<\/strong>. Initially, recruitment will begin in the United States and later expand globally.<\/p>\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-e4eabb8 elementor-widget elementor-widget-text-editor\" data-id=\"e4eabb8\" data-element_type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t<p data-start=\"3158\" data-end=\"3507\">The results obtained to date suggest that AGTC-501 may soon become the <strong data-start=\"3229\" data-end=\"3282\">first gene therapy specifically approved for XLRP<\/strong>. These findings, along with those from the VISTA study, will support the submission of a <strong data-start=\"3372\" data-end=\"3411\">Biologics License Application (BLA)<\/strong> in the United States and a <strong data-start=\"3439\" data-end=\"3484\">Marketing Authorization Application (MAA)<\/strong> in the European Union.<\/p>\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/section>\n\t\t\t\t<section class=\"elementor-section elementor-top-section elementor-element elementor-element-1f7bb7c elementor-section-boxed elementor-section-height-default elementor-section-height-default\" data-id=\"1f7bb7c\" data-element_type=\"section\">\n\t\t\t\t\t\t<div class=\"elementor-container elementor-column-gap-default\">\n\t\t\t\t\t<div class=\"elementor-column elementor-col-100 elementor-top-column elementor-element elementor-element-403d1f4\" data-id=\"403d1f4\" data-element_type=\"column\">\n\t\t\t<div class=\"elementor-widget-wrap elementor-element-populated\">\n\t\t\t\t\t\t<div class=\"elementor-element elementor-element-6e554e6 elementor-widget elementor-widget-text-editor\" data-id=\"6e554e6\" data-element_type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t<p data-start=\"3509\" data-end=\"4018\">The approval of AGTC-501 could <strong data-start=\"3540\" data-end=\"3571\">revolutionize the prognosis<\/strong> for patients with X-linked retinitis pigmentosa, allowing for a meaningful degree of <strong data-start=\"3657\" data-end=\"3676\">visual recovery<\/strong> or at least <strong data-start=\"3689\" data-end=\"3714\">maintenance of vision<\/strong> in treated individuals.
